Proteinase K is a stable and highly reactive serine protease. Evidence from crystal and molecular structure studies indicates the enzyme belongs to the subtilisin family with an active-site catalytic triad (Asp39-His69-Ser224). It is stable in a broad range of environments:pH, buffer salts, detergents (SDS), and temperature. In the presence of 0.1-0.5% SDS, proteinase K retains activity and will digest a variety of proteins and nucleases in DNA preparations without compromising the integrity of the isolated DNA.

Carboxypeptidase Y (CPY) catalyzes the following reaction: Peptidyl-L-amino acid + H2O ——> Peptide + L-amino acid.
It is prepared according to the method of Moore & Stein (J. Biol Chem, 211, 907, 1954). It resembles Carboxypeptidase A in its substrate specificity, but it hydrolyzes C-terminal glycine and L-leuicine more rapidly and L-phenylalanine more slowly.

Translocase Introduction
The enzyme classification and nomenclature list was first approved by the International Union of Biochemistry in 1961. Six enzyme classes have been recognized based on the type of chemical reaction catalyzed, including oxidoreductases (EC 1), transferases (EC 2), hydrolases (EC 3), lyases (EC 4), isomerases (EC 5) and ligases (EC 6). However, it was not noticed back at the time that none of these classes clearly describe an important group of enzymes, which move ions or molecules across membranes or their separation within membranes. Although the hydrolytic reaction is not the primary function, several of these enzymes are involved the hydrolysis of ATP had been previously classified as ATPases (EC 3.6.3.-). Nowadays, these enzymes have been classified under a new EC class of translocases (EC 7).

Chymotrypsin
Background: Chymotrypsin is a digestive enzyme component of pancreatic juice acting in the duodenum where it performs proteolysis, the breakdown of proteins and polypeptides. Chymotrypsin preferentially cleaves peptide amide bonds where the carboxyl side of the amide bond (the P1 position) is a large hydrophobic amino acid (tyrosine, tryptophan, and phenylalanine). These amino acids contain an aromatic ring in their sidechain that fits into a ‘hydrophobic pocket’ (the S1 position) of the enzyme. It is activated in the presence of trypsin.
